“New skin cancer therapy shows promising results”

An advanced therapy for skin cancer that merges immunotherapy with mRNA vaccine technology has shown positive outcomes in a recent advanced trial, as stated in a joint news release by pharmaceutical firms Merck and Moderna on Wednesday. This new treatment, named intismeran, outperformed immunotherapy by itself in lessening melanoma recurrence and containing cancer spread in patients who had previously undergone melanoma surgery.

The treatment’s announcement highlighted “statistically significant and clinically meaningful improvements.” However, doctors remain cautiously optimistic as the Phase 3 trial, the final stage before regulatory approval, is still ongoing, and the disclosed results are from an interim analysis. The companies plan to share the full data at an upcoming oncology conference and with regulatory agencies.

Dr. Marcus Butler, a medical oncologist at Princess Margaret Cancer Centre in Toronto, expressed excitement about the news, noting that positive results from a smaller Phase 2 study indicate potential benefits in the larger Phase 3 trial. Dr. Ramy Saleh, an oncology associate professor at McGill University in Montreal, estimated that if the final trial results confirm the current findings, the treatment may be available to Canadians in over eight months.

The treatment, a combination of Merck’s pembrolizumab immunotherapy drug (Keytruda) and a personalized mRNA vaccine from Moderna, leverages the immune-stimulating properties of immunotherapy and the instructive capabilities of mRNA vaccines to help the immune system target cancer cells based on tumor-specific mutations. Dr. Lennard Lee, a medical oncologist and professor at Oxford, explained that the mRNA vaccine trains the immune system to identify and attack cancer cells by recognizing abnormal proteins found in the tumor samples.

While the trial involved 1,137 high-risk patients with surgically removed Stage IIB-IV melanoma, concerns exist about the potential immune stimulation caused by mRNA vaccines, impacting the interpretation of the trial results. The study, focusing on early-stage melanoma patients, might not directly distinguish the effects of general mRNA immune stimulation from those specific to the melanoma vaccine. Future trials are needed to determine the efficacy of this approach in later-stage melanoma patients.

Melanoma remains a significant health concern in Canada, with around 11,300 new cases expected this year, leading to an estimated 1,250 deaths. Melanoma Canada CEO Falyn Katz emphasized the importance of advancements like intismeran in reducing melanoma-related fatalities.
